چکیده

Myelin is a specialized membrane enriched in glycosphingolipids and cholesterol that contains a restricted set of proteins. The mechanisms by which oligodendrocytes target myelin components to myelin are not known. To identify the sorting determinants for protein transport to myelin, we used a primary oligodendrocyte culture system in which terminal differentiation is synchronized and there is excessive deposition of myelin-like membranes (MLMs). Because several myelin proteins are palmitoylated, we explored the role of acylation in protein transport to MLMs. We found that palmitoylation-deficient mutants of a major myelin protein, proteolipid protein (PLP/DM20), were less efficiently targeted to MLMs. The N-terminal 13 amino acids of PLP/DM20, which are palmitoylated at three sites, were sufficient to direct a fluorescent fusion protein to MLMs. Mutagenesis of the N-terminal palmitoylation motif abolished the transport of the fusion protein to MLMs, indicating that palmitoylation is required for sorting to myelin. Similar results were obtained in myelinating co-cultures of oligodendrocytes and neurons. Furthermore, the combined farnesylation/palmitoylation signals from c-Ha-Ras and the N-terminal consensus sequence for dual palmitoylation from neuromodulin were sufficient for the transport of fluorescent fusion proteins to MLMs. Thus, we conclude that palmitoylation is a sorting determinant for transport to the myelin membrane.

Interplay between phosphorylation and palmitoylation mediates plasma membrane targeting and sorting of GAP43

Phosphorylation and lipidation provide posttranslational mechanisms that contribute to the distribution of cytosolic proteins in growing nerve cells. The growth-associated protein GAP43 is susceptible to both phosphorylation and S-palmitoylation and is enriched in the tips of extending neurites. However, how phosphorylation and lipidation interplay to mediate sorting of GAP43 is unclear. Using ...

Protein palmitoylation in membrane trafficking

Introduction Protein palmitoylation was first described more than 20 years ago for the myelin proteolipid protein, but the functional significance of this post-translational modification remains obscure. Progress has been hampered by the lack of specific inhibitors for acylation and because of the difficulty of developing an assay to isolate the palmitoylating enzyme(s). Palmitoylation generall...

Intracellular sorting of neuromodulin (GAP-43) mutants modified in the membrane targeting domain.

Neuromodulin (GAP-43) is a neurospecific calmodulin binding protein that is targeted to neuronal growth cones via fast axonal transport by an undefined mechanism. The protein is associated with membranes by palmitoylation of cys-3 and cys-4. The objective of this study was to examine the intracellular localization of neuromodulin and neuromodulin mutants modified in the membrane targeting domai...

The role of palmitoylation and transmembrane domain in sorting of transmembrane adaptor proteins.

Plasma membrane proteins synthesised at the endoplasmic reticulum are delivered to the cell surface via sorting pathways. Hydrophobic mismatch theory based on the length of the transmembrane domain (TMD) dominates discussion about determinants required for protein sorting to the plasma membrane. Transmembrane adaptor proteins (TRAP) are involved in signalling events which take place at the plas...

Sorting signals and regulation of cognate basolateral trafficking in myelin biogenesis.

A detailed understanding of trafficking pathways in mature oligodendrocytes is essential for addressing issues aimed at controlling (re)myelination by modulating myelin-directed transport. Previously, we have shown that viral marker proteins HA and VSV G, on reaching the apical and basolateral surfaces of polarized epithelial cells, respectively, are primarily transported to the plasma membrane...
